Principle : Trizol (Total RNA Isolation Reagent) is a ready-to-use reagent for the isolation of total RNA from cells
and tissues for use in PCR analysis. Trizol is a monophasic solution of phenol and guanidine
isothiocyanate. During tissue homogenization or lysis, the Trizol maintains RNA integrity, while
disrupting cells and dissolving cell components. Addition of chloroform followed by centrifugation
separates the solution into an aqueous phase and an organic phase. RNA remains in the aqueous phase
and recovered by precipitation with isopropanol. The RNA redissolved in Nuclease-free water. RNA is
stored for research requests.
Warnings/Precautions
A. Use Standard Precautions when handling all body fluids, tissues and cell cultures. Refer to the
Specimen Collection.
B. Trizol is toxic when in contact with skin and if swallowed. It will cause burns. Be sure to wear a lab
coat, gloves and safety glasses when working with Trizol. If in contact with skin, wash immediately with
plenty of soap and water.
C. To prevent RNase contamination, always wear gloves. Use sterile, disposable plastic ware and
pipettes dedicated strictly to RNA work to prevent cross-contamination with RNases from shared
equipment
Reagents
Chloroform, DEPC Water, 75% Ethanol, Isopropanol.
Starting Materials:
Cell in Monolayer/suspension Culture: Up to 1×۱۰۷ cells, depending on the cell line.
Bacterial cells: Up to 1×۱۰۷ cells
Tissue: Up to 100mg
Plant and Fungi: Up to 100mg
Liquid materials (like serum): 100μl
Whole blood: 500μl (up to 2ml)
